Isolation of pseudomonas aeruginosa from hospital infections and investigation of surface virulance factor (BLAIMP) by molecular method

Pseudomonas aeruginosa has become a global threat due to its increasing resistance to existing antibiotics. In this study, it was aimed to retrospectively determine the antimicrobial resistance of P.aeruginosa isolates isolated from different sources. Among the 150 isolates we collected, The samples were taken from patients attended Al-Mosul Governmental Teaching Hospitals . Samples were diagnosed based on cultured characteristics on macConkey agar medium consider selective medium, microscopic examination by observing the rod-shaped bacterium, and biochemical tests such as catalase (+), gram (-), citrate (+).To increase accuracy in the diagnostic process . We got 21(14%) P. aeruginosa from different sources included clinical source 8(38%) and environmental source 13(61.9%) . Among 21 samples were only 2(10%) sample of P. aeruginosa were resistance to Imipenem and the activity of responsible gene(blaimp) for resistance phenomenon was determined using the real-time technique by RNA isolation from 21 sample ,cDNA synthesis and gene expression using specialized gene primers. Finally, two samples among all resistance strains showed significance blaimp gene expression up regulation, whereas the other remaining strains revealed non significance gene activity and the results were respectively 6,995424, 2,876129
