Investigation of the effect of primula vulgaris extract loaded silver nanoparticles on BCL2, MDR1 and P53 gene expression levels in hela cell lines

Cervical cancer is the second most common type of cancer among women worldwide after breast cancer. HeLa cells are a human cervical cancer cell line. HeLa is the most widely used model cell line to study the molecular biological properties of cell lines cancerous cells. In this study, it was aimed to investigate the effects of flower and leaf extracts loaded silver nanoparticles (AgNP) of Primula vulgaris plant on cell proliferation and BCL2, MDR1 and p53 gene expression levels in HeLa cell line. AgNP IC50 dose values of Primula vulgaris plant loaded with flower and leaf extracts were determined by iCELLigence real time cell analysis system. After IC50 dose determination, IC50 dose group (1st Group), LD50 dose group (2nd Group), Cisplatin added group (3rd Group), AgNP dose group without plant extract (4th Group) and control group (5th Group) ), BCL2, MDR1 and p53 gene expression levels in 5 different groups were analyzed by qRT-PCR method. To HeLa cell lines; After the addition of AgNP loaded with Primula vulgaris flower extract, the IC50 values determined at the 12th hour and 24th hour are 58,39 ug/mL and 57,65 ug/mL, respectively (r2 = 1). Following the addition of AgNP loaded with Primula vulgaris leaf extract, the IC50 value determined at the 12th hour and 24th hour is 42,14 ug/mL and 48,50 ug/mL, respectively (r2 = 1). It was determined that the death rate of HeLa cells loaded with Primula vulgaris flower and leaf extract loaded with AgNP increased depending on the dose (p<0.05). According to the control group in the AgNP dose group loaded with Primula vulgaris extract; A 0.8-fold decrease in the amount of BCL2 gene expression, a 1.1-fold decrease in the MDR1 gene expression amount and a 1.3-fold decrease in the amount of p53 gene expression were observed. As a result, it has been determined that Primula vulgaris plant inhibits the proliferation of HeLa cells. Further studies are needed to test the clinical applicability of the study results.
