Abstract (EN)
Cancer is one of the most important health problems of our era. It is also a public health problem due to its high incidence and high mortality rate. Changing living and nutritional conditions, high radiation and most importantly genetic predisposition are some of the causes of cancer cases. Chemotherapy is a method used in cancer treatment. It means treating the tumor with medication and chemotherapy; damages cancer cells, prevents from the growth and proliferation of cancer cells, which are capable of unlimited division. Doxorubicin, sold under the trade name Adriamycin, is a chemotherapeutic. Doxorubicin, an anthracycline derivative antibiotic; shows its effect by intercalating into DNA like all other anthracyclines. Although the exact mechanism of action of doxorubicin is still not fully understood, it is known that intercalation with DNA stops replication. Doxorubisin stabilizes the DNA Topoisomerase II complex, which enables the DNA double helix to open during DNA replication, and inhibits replication. Doxorubicin is frequently used in the treatment of various cancers such as hematological cancers, various types of carcinoma, and soft tissue sarcomas. Piperlongumin is an alkaloid natural product found in Piperlongum Linn plant species and showing anti-tumorigenic activity in cancerous cell lines and animal models. Many studies suggest that piperlongum may be a chemopreventive agent. In addition to studies suggesting that it achieves its anti-cancer effect by silencing the GSTP1 and / or GSTO1 genes, there are studies suggesting that it prevents cancer cell migration and therefore metastasis. Within the scope of this completed Master's Thesis, the comparative expression levels of genes associated with metastasis and tumorigenesis such as MMP9, PTEN and NOTCH1 in the cervical cancer cell line HeLa cell line of doxorubicin and piperlongumin were evaluated by Real Time PCR and the expressions of the proteins encoded by these genes by Western- Blot method. Within the scope of this completed Master's Thesis, the comparative expression levels of genes related to metastasis and tumorigenesis such as MMP9, PTEN and NOTCH1 in the cervical cancer cell line HeLa administered doxorubicin and piperlongumin were evaluated using Real Time PCR and the expressions of the proteins encoded by these genes by Western- Blot method.
