Isolation and characterization of probiotic bacteria from the plant Herniaria glabra L. and determination of the in vitro wound healing properties

Discovery of new probiotic candidates is therapeutically important because of their superior properties such as probiotics, immunomodulators, anticarcinogens, anti-infectives, and pathogen suppressors. In this context, it was aimed to identify new probiotic candidates in this study. H. glabra plant was preferred to obtain probiotic candidate bacteria. First of all, bacterial isolation from the plant and identification of isolated bacteria at molecular level by 16s rRNA sequencing, then probiotic characterization of preferred bacteria and their wound healing effect were analyzed by cell culture studies. As a result of these studies, the characterization of two important lactic acid bacteria [Enterococcus mundtii (E. mundtii) AF1 and Pediococcus pentosaceus (P. pentosaceus) AF2] was performed. It has been determined that both bacteria are quite resistant to gastrointestinal system (GIS) conditions. In the safety tests, it was determined that two bacteria had γ-hemolytic activity and were generally sensitive to the tested antibiotics. The ability of bacterial supernatants to contain antimicrobial metabolites was evaluated by agar well diffusion method, and E. mundtii bacteria against B. subtilis, K. pneumoniae and Y. pseudotuberculosis bacteria, and P. pentosaceus bacteria E. coli, B. subtilis, P. vulgaris, S. typhimurium, P. aeruginosa, K. pneumoniae, E. cloacae and Y. pseudotuberculosis bacteria were determined to show inhibition activity. Autoaggregation values were calculated as 73.44 % for P. pentosaceus and 72,46 % for E. mundtii. Coaggregation values of P. pentosaceus sample against E. coli and L. monocytogenes bacteria were calculated as 56,8 % and 57,38 %, and E. mundtii sample as 70,77 % and 63,78 %. Finally, the healing effect of isolates on the wound formed in vitro in cell culture medium (L929 cell line) was tested and it was determined that probiotic candidate bacteria showed wound closure promoting healing activity. This study revealed that the use of E. mundtii AF1 and P. pentosaceus AF2 bacteria as potent probiotic candidates can be evaluated.
