Abstract (EN)
In this thesis, two hydrazinated chalcone derivative pyrazoline isomer compounds (4.2 / 2.2) are investigated as suitable fluorescence probes for biological systems. For this purpose, interactions of 4,2 and 2,2 isomers with Sodium Dodecyl Sulfate (SDS), which is accepted as in-vitro model membrane system, and human serum albumin (HSA), which is the model carrier protein, were investigated using absorption and fluorescence spectroscopy technique. The study mainly includes the investigation of fluorescence energy transfer in the aforementioned environments according to Förster's theory and Stern-Volmer equation. For this purpose, while 4, 2 and 2, 2 pyrazoline derivative molecules are used as donors in SDS micelle medium, Safranin T, Akridin O, Pyronin Y, and Floresin molecules were preferred as acceptor molecules. In the HSA environment, while the donor molecule is HSA, acceptor 4, 2 and 2, 2 are isomer compounds. Energy transfer parameters and Stern-Volmer parameters were calculated for these donor-acceptor pairs, whose spectral overlap areas were suitable, and the compatibility of donor-acceptor pairs was supported by the calculated parameters. Energy transfers in the SDS and HSA system, Stern-Volmer, and thermodynamic parameters expressed the compatibility of molecules for biological systems. Also, to determine whether the molecules are suitable for biological system studies, whether they are cytotoxic was investigated by in-vitro cancer cell line studies and both molecules were found to be non-toxic. To understand whether these molecules can be used in cell imaging techniques, confocal microscope images were obtained by in-vitro cell application. As a result of this study, it was understood that the molecules stain the cell cytoplasm, so both molecules can be used for cell staining at 360 nm wavelength in cell imaging processes.
