Investigation of probiotics properties of lactobacillus species isolated from turkeys

Abstract (EN)

Objective: The object of this research is to analyse probiotic functions of isolated Lactobacillus spp. from ileum and contribution to growth of turkeys. Material and Methods: In November and December months of 2021, 50 numbers of ileum contents that was taken from a custom poultry slaughterhouse was given to Adnan Menderes University's Faculty of Veterinary Medicine's laboratory via cold chain. The samples were inoculated to the selective mediums and incubated. As Markiewicz ve Biedrzycka (2005) used to do it before, special PCR technique is used for the identification of genus of Lactobacillus spp. to test the bile resistivity of isolated bacteria, bile tolerence test was made. With pH tolerence test, resilience of isolated bacterium were identified. Samples were inoculated to Congo red MRS medium for searching of hydrofobicities. The sample's antibiotic resistency mechanisms were investigated with Kirby Bauer Disk Diffusion technique. Results: As a result of 50 ileum contents which are taken from approaching slaughter aged turkeys, inoculated to specific medium under 35°C during 48 hours. Only 27 of them have seen bacterial reproduction. After PCR, these 27 isolates' only 6 of them just identified as Lactobacillus acidophilus. By bile tolerance test, only 3 isolates were seen % 100 resistant to % 0,5 bile concentration. In pH tolerance test, only 2 isolates that have been exposed to pH 5 solutions were % 100 resistant. All Lactobacillus spp. isolates' only 7 of them and two of Lactobacillus acidophilus isolates were identified as non-hydrophobic. As the conclusion of Kirby Bauer Disk Diffusion antibiogram technique; Lactobacillus spp. isolates were indicated as highly resistant to three antibiotics but highly sensitive to one of them. Lactobacillus acidophilus isolates were indicated as highly resistant to three antibiotics but highly sensitive to four of them. Conclusion: As a result of this study; vitality of probiotics were observed as getting decreased if the concentration of bile is increased. But if, pH value is became getting neutral, the vitality of probiotics were getting increased. This research shown that usage isolates of Lactobacillus acidophilus and Lactobacillus spp. which are resistant to multiple antibiotics, can be used as probiotics and stimulate the growth of digestive tract of turkeys.
