Glutamate oxidase enzyme production from Hypocrea Jecorina pure culture

L-glutamate oxidase (L-glutamate oxidase EC 1.4.3.11)is the important enzyme that catalyzes the deamination of L-glutamate to alpha - ketoglutarate and ammonium ions. It has an important role in the nitrogen metabolism. Recently, L-glutamate oxidase has received much attention with respect to its therapeutic and industrial applications . Therapeutically in many of liver and heart diseases effects in metabolism are effected with this enzyme activities. Also in mass food production industry the content of mono sodium glutamate (MSG) had very important role in Alzheimer diseases. In this case to analysing the amount of MSG with this enzyme's activity and for biosensor research this study is highlighted. Glutamate oxidase activity is widely distributed in plants , animal tissues and microorganisms including bacteria, yeast and fungi. It has aimed that microbial production of L-glutamate oxidase enzyme from Hypocrea jecorina pure culture, determination of optimum conditions and calculation of kinetic parameters of these produced enzymes. As a result of work, it was determined optimum pH value as 8.5 and optimum temperature as 37-40oC for L-glutamate oxidase enzyme produced microorganism. Km and Vmax values, the kinetic parameters of enzyme produced Hypocrea jecorina pure culture , was determined as 2.58 mM, Km value and, 83.33 U/mg protein of Vmax value by plotted Lineawaever-Burk graph, respectively. In addition, thermal and storage stability of L-glutamate oxidase enzyme produced from microorganism specy was determined as very high.
