Özet (EN)
The female reproductive tract occurs ovary, fallopian tubes, cervix and uterus. Although oocytes and sperm morphology is important for successful implantation process, fertilization take place smoothly and developing embryo in the fallopian tube to the morula stage after coming up is required to reach the site of implantation. After all of these steps take place, the receptivity of the endometrium and embryos adhesiveness stand outs for implantation. Cause of unexplained infertility cases especially endometrial problems are seen to be more. İmplantation process is controlled with endometrium, factors secreted by the embryos and in accordance with these factors embryo and/or endometrium via receptors on. Although fertilization and early embryonic development are developing normally, when there isn't endometrial acceptability, implantation can not occur, so advanced embryonic development could not take place. MicroRNA (miRNAs) are small noncoding RNAs that control post-transcriptional gene expression. More than 700 human miRNAs have been identified and they were shown to play an important role in intracelluler cycle regulation in both normal and pathological conditions. With recent studies in the female reproductive system organs at different periods have been shown to be different expressions of different miRNAs, and their normal regulation and their distribution of abnormal cases and roles have begun to be explored. In this study we aim to idetified miRNAs that is expressed in endometrial biopsy samples which are taken proliferation and early secretion periods in fertile and infertile cases, and also analyses their distribution in infertile cases with implantation process. For this purpose, we take the samples from fertile and infertile cases in proliferation and early secretion periods. Women which we selected, are between the ages of 25-38 and they have applied to Celal Bayar University Hospital IVF Center and Hafsa Sultan Hospital in Obstetrics and Gynecology clinic. Some of these samples will be taken up in solution in 10% formalin for histological analysis and the remaining samples directly put into liquid nitrogen for miRNA analysis that will be kept. In histological analyses, the samples are painted with hematoxylen-eosin for morphological analysis after the routine parafin tracking and for the immunohistochemical analysis anti-ago1, anti-dicer, anti-drosha, anti-eIf2 distribution will be looked. For the miRNA analyses, after RNA extraction in the samples, RNA isolation and quality will be analyzed with quantitative real time polymeraz chain reaction (qRT-PCR), and miRNAs that have identified, are miR-17-5p, miR-23a, miR-23b, miR-542-3p, miR-21, miR-199a*, miR-705, miR-20a, miR-26a, miR-125b, miR-200a/b/c.
