Investigation of the effects of dosetaxel and abiraterone acetate on notch1, jagged1 and hes1 expressi̇ons in human prostate cancer cells in vitro conditions

Abstract (EN)

Objective: Prostate cancer (PCa) is the most common type of cancer observed in men. In men, it ranks second in cancer-related deaths. There have been no studies yet in the literature about how the expression of Notch signaling pathway members, an important pathway in cell proliferation, differentiation and fate are affected by docetaxel and abiraterone acetate used in PCa treatment, and how these expressions will affect the treatment process. Therefore, the aim of this study was to investigate the effects of abiraterone acetate and docetaxel on the expression of Notch1, Jagged1 and Hes1 in prostate cancer cell lines. Method: After LNCaP and PC3 cell lines were cultured at 37 ºC for 72 h in vitro, the effects of abiraterone acetate and docetaxel on Notch1, Jagged1 and Hes1 expression were examined by MTT proliferation assay, immunoflourescence, western blot and qRT-PCR. Results: According to the findings obtained from our study; while abiraterone acetate, docetaxel and abiraterone acetate + docetaxel to PC3 cells was decreasing the level of Notch1, especially increased the level of mRNA for Hes1 at very important level. Abiraterone acetate and abiraterone acetate + docetaxel reduced Notch1 expression in LNCaP cells. In LNCaP, Jagged1 expression was not significantly affected by abiraterone acetate, docetaxel and abiraterone acetate + docetaxel administration. Hes1 expression was significantly increased in both gene and protein level groups at all drug groups. Conclusion: When abiraterone acetate and docetaxel were administered to prostate cancer cells in vitro, the expression of Notch signaling pathway proteins changed. With the application of these drugs; in both cell lines, especially Hes1 expression showed a very significant increase. Therefore, co-administration of Notch signaling pathway inhibitors together with these chemotherapeutic drugs in the treatment of prostate cancer patients, It was thought that Hes1 expression could be stopped the deterioration of the prognosis of the patient.
