Antitumoral effectiveness of cow, sheep, goat and donkey colostrum on a549 and mcf-7 different cancer cells

1.2. Abstract Aim: Colostrum is a natural compound containing antibodies/immunoglobulins that the offspring will receive from the mother during the first month of life. In this study, it was aimed to investigate the effectiveness of whey and casein proteins obtained from colostrum of different species such as cow, sheep, goat and donkey on human lung cancer cells (A549) and breast cancer cells (MCF7). Material and Method: Colostrums were fractionated into whey and casein proteins. These proteins were determined by SDS-PAGE electrophoresis and sterilized with a membrane filter. Antitumoral activity levels of different concentrations of lyophilized proteins were measured by MTT viability test on cells and IC50 values were determined. In addition, cytokine levels (IL-2, IL-6 and TNF-α) in colostrums were measured. Results: Serial dilutions of whey and casein proteins in colostrum starting with 6400 µg/ml were incubated with A549 and MCF-7 cells for 24, 48 and 72 hours. When cow whey proteins were incubated with MCF-7 cells, cell viability and IC50 values were determined as 37.06% and 3144 µg/ml, respectively. Cell viability and IC50 of donkey whey proteins after MCF-7 cell incubation were measured as 16.64% and 3424 µg/ML, respectively. In A549 cell incubation of sheep casein, cell viability was 29.38% and IC50 was 3612 µg/ml; while cell viability was 37% and IC50 was 3383 µg/ml in MCF-7 cell incubation of sheep casein. Conclusion: It was concluded that cow and donkey whey and sheep casein proteins have antiproliferative and anticarcinogenic effects on A549 and MCF-7 cancer cell lines.
