Abstract (EN)

ABSTRACT Investigation of the Relationship Between the Results of the Morphometric Analysis of Granulosa Cells Surrounding the Oocytes of the Patients Admitted to the In Vitro Fertilization Unit due to Infertility and Fertilization Aim: Infertility is a serious problem that is seen in about 15% of the couples of reproductive age on the world. It is noteworthy that female infertility rate is high in classification of infertility. İn vitro fertilization (IVF) treatment that is one of the assisted reproductive techniques (ART) and applied for the purpose of infertility treatment has become a source of hope for couples that have no children. The ovulation disorders, uterine, tubal and cervical factors are found among the causes of female infertility. In addition, female age, oocyte, and embryo quality are also other factors affecting fertilization and implantation. In this study, it was aimed to investigate the relationship among the patient age, FSH, LH, TSH, T3, T4 and Ca+2 levels by making the morphometric analysis of granulosa cells around the oocytes of the patients undergoing IVF treatment. Material and Method: Fifty infertile women patients were randomly selected among the patients who applied for IVF treatment to the IVF Unit of the Ataturk University Research Hospital, and two groups were divided as under the age of 35 and over the age of 35. Granulosa cells in the cumulus oocyte complex taken from these patients during oosit pick-up (OPU) procedure were used. Morphometric measurements of granulosa cells stained with "Papanicolaou" staining method were performed under light microscope. Results: The age of the patients was evaluated together with beta-HCG, FSH, LH, TSH, T3, T4 and Ca+2 values. The obtained data were analyzed statistically. When the measurements of the cytoplasm and nucleus areas of granulosa cells were compared with biochemical parameters such as beta-HCG, FSH, LH, TSH, T3, T4 and Ca+2 values, it has been observed that there is no significant difference between the groups of under the age of 35 and over the age of 35. At the end of further statistical analyzes, it was determined that there was a significant correlation between age and TSH, between the cytoplasm area of granulosa cells and serum T4 levels, between serum FSH levels and Ca+2 values, and also between serum T4 and Ca+2 values. Conclusion: According to our findings, it was concluded that in addition to FSH, LH, TSH, T3 and T4, Ca+ 2 could affect oocyte quality and fertility.
