Investigation of the effects of moringa oleifera on the AKT/FOXO3A/bim path in human glioblastoma cell lines

Glioblastoma, which is the most common glioma among gliomas, is still considered as stage 4 by WHO and is a mortal tumor despite current treatments. Moringa oleifera is an herb whose anticancer and apoptosis-inducing effects have been discovered on the common tumorogenesis pathway with glioblastoma in many cancer types. In this study, the effect of Moringa oleifera on the AKT/FOXO3a/Bim pathway known in the pathogenesis of glioblastoma was investigated to contribute to the treatment options. U87-MG and T98-G glioblastoma cells were cultured in vitro. Moringa oleifera was applied to U87-MG and T98-G cell lines for cytotoxic activity detection. Immunocytochemical optical density measurements were compared in U87-MG and T98-G control and experimental groups using Image J software. In addition, colony formation percentages in U87-MG and T98-G control and experimental groups were evaluated with Colony Count J software. The decrease in AKT and the increase in FOXO3a and Bim were found statistically significant in the U87-MG and T98-G experimental groups compared to the control group. The decrease in colony formation percentages was also significant in the U87-MG and T98-G experimental groups compared to the control group. As a result, it was determined that Moringa oleifera triggered apoptosis in U87-MG and T98-G glioblastoma cell lines via the AKT/FOXO3a/Bim pathway.
