Abstract (EN)
In recent years, biopolymers are widely used in various fields of medicine; namely, orthopedics, chemotherapy, immunology and dentistry. It is very important to test compatibility to determine the practical applicability of any polymer. Many effects of polyacrylic acid (PAA), which is a polyelectrolyte, is known by researchers. Its adjuvant effect in immunology, antitromogenic effect in medical experiments and its use as transporter in drug release systems is among its applications. Despite there are various studies of PAA with several different conjugates, there is no study on its individual toxicity. It is severely important to determine the toxicity in biopolymer studies and several cell lines are used for this kind of studies. But there is no information on the sensitive cell lines for PAA and its conjugates. The aim of this thesis is to make the culture of human mammary cancer cell line MCF-7 and mouse fibroblast cell line L-929 and examine the PAA toxicity on these cells by using various methods. The methods used in this study are; cryopreservation, 3-(4,5-dimethyltriazol-2-il)- 2,5-diphenyltetrazolium bromide (MTT), trypan blue and 4,6-diamidino 2 phenylindole (DAPI). Initially, the adaptation of MCF 7 and L-929 cell lines is provided in the tissue culture laboratory of Yıldız Technical University, Department of Bioengineering for the first time and their stable cultures were established. Optimization of cryopreservation conditions was achieved and satisfactory cryobanks of these cells were generated in the laboratory. In this study, it is established for the first time that, MCF 7 cell line is a suitable cell line for toxicity studies of PAA. In the latter steps of culture studies which include toxicity analysis of PAA in varying pH and concentration; differential toxicity in different cell lines was detected. IC50 value of PAA is shown to be 6,6 in MCF 7 cells whereas 1,8 in L-929 cell lines. These results indicate the importance of the choice of cell lines used in toxicity analysis.
