Investigation of the effect of different particle size allografts on human osteoblast cell cultures with resveratrol incubation

Abstract (EN)
This study aims to examine in vitro how osteoblast cells are affected by allograft biomaterials of different particle sizes used for defects in maxillofacial surgery, together with resveratrol that is thought to have a positive effect on bone development and cell proliferation. For examinations 7 main groups and 21 sub-groups were organized according to 24-, 48-, and 72-hour intervals. These are; the first control group only osteoblast culture was added to petri dishes (A), the second control group, only 0.02% DMSO, a resveratrol solvent, was added to osteoblast culture (B), the osteoblast culture containing incubated resveratrol in addition to DMSO without bone grafts (C), the osteoblast culture containing a small-particle bone allograft with incubated resveratrol in addition to DMSO (D), the osteoblast culture containing a large-particle bone allograft with incubated resveratrol in addition to DMSO (E), the osteoblast culture containing only a small-particle bone allograft (F), the osteoblast culture containing only a large-particle bone allograft (G). ACC 243 (osteoblast cell line based on osteosarcoma of human origin) was used as an osteoblastic cell line. Resveratrol dissolved in 0.02% DMSO was used at a final concentration amount that was determined to be 2 μM. WST-1 test kit was used to evaluate the cytotoxicity and also to determinate the cell vitality and proliferation. The IBM SPSS Statistics 2.2 (IBM SPSS, Turkey) software program was used in the evaluation of statistical analyses of the findings obtained in the current study. The comparison of the control group B with the group C and group E were found statistically significant according to all time intervals while the comparison with group D was found statistically significant only at the end of 72 hours. The comparison of the group G that was treated with large particle sized allografts with the group C, D and E were found statistically significant according to all time intervals. According to the results of our study, resveratrol had a negative effect on osteoblastic activity when applied together with allograft materials. We believe that more comprehensive studies in the future will contribute to the assessment of being a material that is feasible for clinical practice.
