Evaluation the effect of some plant extracts on gene expression of OPRL in Pseudomonas aeruginosa from Iraqi patients

The current study included a collection of (150 ) swabs from patients suffering from burns and wound infections in both genders, for the period from March 2022 to June 2022, from injured patients whom attending to Baghdad Teaching Hospital, Ghazi Hariri Hospital, Central Laboratories in Medical City and Al-Yarmok hospital in Baghdad /Iraq. After final diagnosis of samples, among 130 positive culture samples, only 32.3%% (42/130) isolates belonged to P. aeruginosa, and 67.7% (88/130) isolates belonged to other genera of bacteria, Identification of P. aeruginosa was done by Microscopic ,Biochemical tests ,Cetrimide agar , Kliger Iron Agar (KIA). The susceptibility test of bacterial isolates against 11 selected antibiotics was conducted by VITIK 2which indicates that the P. aeruginosa isolates are fully resistance 42 (100%) to 3 of the antibiotics of interest during the current study, particularly Cefazolin, Ceftriaxone and Trimethoprim/Sulfamethoxazole. Meropenem was the most effective antimicrobial agents against P. aeruginosa isolates because P. aeruginosa, was showed high percentage of sensitivity to it 36 (85.71%), followed by Ciprofloxacin 28(66.66%) and Piperacillin /Tazobactam 26 (61.90%). Detection of Antimicrobial Activity of Curcuma longa plant extract by Well agar diffusion method in the present study, of 42 isolates of P. aeruginosa against plant extract was studied to determine the pattern of isolates resistance based on the diameter of the region around the antimicrobial well and compared with the standard diameters of the antibiotic gentamycin, the vast majority of P. aeruginosa isolates were sensitive to turmeric extract by 59.5% (25/42), and the isolates that had resistant and Intermediate resistant to Curcuma extract they constituted 19.0% and 21.4%, respectively, of the P. aeruginosa total isolates study. The bacterial DNA, and RNA was extracted from isolates of P. aeruginosa. PCR was carried out for all bacterial isolates, using specific primers, to detect OPRL gene. The results showed that the 25 P. aeruginosa isolates have the OPRL gene in a percentage 59% from all isolates. The results also showed expression levels of OPRL gene regarding to the fold change by using RT-PCR, the results showed before Curcuma plant extract 1.5243±0.243, and 4.014±30.12421053 for after Curcuma plant extract with highly significant difference P- value 0.01
