Abstract (EN)

As the side effects of synthetic components increase, consumers are increasingly opting for products containing natural and healthy ingredients. Due to the positive health effects of products derived from waste produced in the food industry and the low cost of processing these wastes, studies on the utilization of food waste products in various fields such as food, dye, pharmaceuticals, and cosmetics have accelerated. This thesis aims to investigate the content and pancreatic lipase inhibitory effects of extracts prepared with experimental design from the pomace of the Isabella grape (Vitis labrusca), an endemic value for the Black Sea Region. The İsabella grape pomace used in this study was lyophilized, ground, and used to prepare extracts according to a fractional factorial design. The phenolic contents of the obtained extracts were determined by RP-HPLC. The parameters that most affected the extraction were found to be ethanol percentage, microwave power, and temperature. Box-Behnken Optimization was used to optimize the extraction conditions. Considering all data, extracts containing high amounts of phenolic substances and anthocyanins were identified as optimal extracts. The study found that the extracts obtained from grape pomace exhibited lipase inhibitory effects, with the optimal extract's lipase inhibitory effect being close to that of Orlistat. To determine the antioxidant capacity of the optimal extracts, CUPRAC, FRAP, and DPPH radical scavenging activity studies were conducted. It is believed that these wastes resulting from the processing of grapes could contribute to the development of food and health products aimed at improving general health and preventing oxidation-related diseases as a natural source of antioxidants; they could also be a potential source for the search for new pharmacophores with lipase inhibitory effects in obesity treatment.
