The relationship of calreticulin outcomes of jak2 positive and JAK2 negative myeloproliferative patients with clinical parameter

Abstract (EN)

Background and aims: Chronic myeloid neoplasms are pluripotent hematopoetic diseases characterized by maturating and differantiating defects of bone marrow cells and reflecting to peripheral blood. In this study, we investigated the relationship of calreticulin outcomes of JAK2 positive and JAK2 negative myeloproliferative patients with clinical parameters Materials and Methods:In this study, we included 194 patients admitted to Gaziantep University Faculty of Medicine between January 2014 and December 2016. Of these patients, 79 were JAK2 positive and 116 were JAK2 negative patients. Normal distribution fitness of numerical data was tested by Shaphiro Wilk test. Student T test was used for comparison of normal dividing variables in two groups and Mann Whitney U test was used for comparison of normal dividing variables in two groups. The Kruskall Wallis test was used to compare the normal non-dispersive variables in 4 groups. The relationship between categorical variables was tested by Chi square test. Relationships between normal non-dispersive numerical variables were tested with Spearman rank correlation coefficient. SPSS 22.0 package program was used in the analyzes. P <0.05 was considered significant. Results: We studied 194 patients and found 2 CALR mutations. There was a significant correlation between JAK2 mutation and thrombosis (p = 0.041). There was no significant correlation between JAK2 mutation and CALR mutation (p = 0,790). For the patients who had thrombosis at the time of diagnosis or before the diagnosis, the laboratory was not related to the diagnosis at the time of diagnosis (p> 0,05). There was no correlation between spleen size and LDH (r = -0,088, p = 0,765). There was no significant correlation between LDH and hepatomegli (p = 1). There was no significant relationship between LDH and itching (p = 0.541). There was no significant relationship between LDH and fibrosis (p = 0,390). There was no significant correlation between CALR mutation and thrombosis (0,744). There was no significant correlation between CALR and hepatomegaly (0,286). There was no significant relationship between CALR and itching (0,406). JAK2 positivity in CPPs has an important role in clinical features and complication development. The ratios we have found in our studies show compliance with the literature. In the literature studies, JAK2 mutation has been studied in 12 regions of exon and JAK2 mutation in our study is not compatible with the literature since it is studied in 14 regions of exon. The CALR mutations in our study are E381A (c.1142A / C), c.1099_1050delL367.
