Abstract (EN)

Zucchini Yellow Mosaic Virus (ZYMV) is one of the major viruses causing economical losses in cucurbit production fields. The aim of this study, biological, serological and molecular detection of ZYMV growing cucurbit in greenhouse and field in Adana and Mersin provinces between 2006 and 2007 and possible using of plant activators in control of ZYMV. Collected samples that suspicious infected virus of plants were firstly tested with ELISA and infection ratio was determined as 51.2% ZYMV. Positive one of them was coded and used for mechanical inoculation and vector transmission. Total RNA extraction was done from ZYMV infected test plants and used for RTPCR method. Morever, plant extract was used for IC-RT-PCR method. DNA bands were observed in respected sizes after the PCR products were tested in agarose gel electrophoresis. The effect of ACTIGARD, MESSENGER and ISR-2000 on viral disease development were determined with pot and field experiments in order to induce resistance against ZYMV on squash. ZYMV inoculation after 48 hours of ACTIGARD applications and 72 hours of MESSENGER and ISR-2000 aplications were observed effective on symptom expression time compared to control plants. Lignification and H 2O2 mass were detected on plant activators treated and SAR developed plants by histochemical staining. Key Words: ZYMV, RT-PCR, Plant Activator, SAR, Histochemical Staining
