Abstract (EN)

Olanzapine (OLA) is an important molecule in medicine and pharmacy used in the treatment of some mental disorders. OLA, which has been the subject of many studies, has been selected as a test substance in this thesis. No study has been found in the literature regarding the determination of OLA using PGE. Therefore, it was aimed to investigate the voltammetric behavior of OLA at PGE and to develop a sensitive, rapid and inexpensive determination method using square wave voltammetry (SWV). The manner of conditioning of PGE (P-trt A, P-trt B), type of supporting electrolyte, pH, scan rate, initial and anodic boundary potential, waiting time and device parameters (pulse amplitude, frequency, potential step) were optimized. Under the optimum conditions, the variation of the OLA peak currents was linearly found from 5.00 ×10-7 to 6.00 ×10-6 M concentration range for P-trt A and from 6.25 ×10-8 to 2.00 ×10-6 M concentration range for P-trt B. Limit of detection and limit of quantification olanzapine for P-trt B were calculated as 4.71 ×10-8 M and 1.57 ×10-7 M, respectively. The developed method was successfully applied to Zyprexa Velotab (5 mg OLA / tablet) in the form of an orodispersible tablet. The application was made with both standard addition (method 1) and calibration graph (method 2) methods and the results were compared with the tablet content. However, since the drug format was different, the preparation of the tablets for analysis was carried out in a manner different from conventional practice in the literature. The sample preparation step not requiring filtration, the performance and cheapness of the electrode used, the sensitivity and simplicity of the method even though there is no complex electrode modification are important advantages of the study.
