Synthesis and antibacterial properties of calix[4]arene based water-soluble silver nanoparticles

Abstract (EN)

This study includes the synthesis of a new calix[4]arene macromolecule containing a water-soluble hydrazone derivative, Then, obtaining silver nanoparticles from this compound in suitable medium, and determination of its antibacterial properties. The structures of the synthesized compounds were elucidated using spectroscopic methods (FT-IR, 1H-NMR, TEM, Elemental analysis). In synthesis studies, firstly, 5,11,17,23-tetra-tert-butyl-25,26,27,28-tetra-hydroxy-calix[4]arene (1) was synthesized using methods known in the literature. In accordance with another method known in the literature, this compound was reacted with methyl bromoacetate in the presence of K2CO3 in the presence of acetone as a solvent, 5,11,17,23-tetra-tert-butyl-25,26,27,28-tetra-(methoxycarbonylmethoxy)-calix[4]arene (2) compound was synthesized. Afterwards, compound numbered (2) is reacted with hydrazine hydrate in the presence of ethyl alcohol as a solvent, and containing amide group 5,11,17,23-tetra-tert-butyl-25,26,27,28-tetra-(carbohydrazide-methoxy)-calix[4] arene (3) was obtained. Compound (3) is a water-soluble 5,11,17,23-tetra-sulfonato-25,26,27,28-tetra-(carbohydrazide-methoxy)-calix[4]arene (4) compound, which is treated with H2SO4 has been obtained. Finally, C[4]SO3H-AgNPs (5) nanoparticles were obtained by activating the aqueous solution of the compound (4) with AgNO3. Pseudomonas aeruginosa ATCC 11778, Escherichia coli ATCC 35213, Staphylococcus aureus ATCC 12600, Streptococcus mutans ATCC 10449 were used as test microorganisms to examine the antibacterial properties of synthesized substances (4), and (5). In accordance with the findings obtained, compound (4) was as effective as the first positive control sample (vancomycin) on Pseudomonas aeruginosa ATCC 11778, showed much more antibacterial effect than the second positive control sample (clindamycin), and on Escherichia coli ATCC 35213, it was more effective than the first positive control sample. (vancomycin) approximately six times more, and again more antibacterial effect than the third positive control sample (ofloxacin), more antibacterial effect on Staphylococcus aureus ATCC 12600 than the first positive control sample (vancomycin) and finally its effect on Streptococcus mutans ATCC 10449, when examined, it was observed that the compound had more antibacterial effects than the first positive control sample (vancomycin).
