Abstract (EN)
Problems on blood collection and storage, excessive costs, reserve deficiency around the world has led to development of the material instead of blood. Artificial blood works base development of the oxygen-carrying compound. The main objective of this work is development of biological material that will be blood alternative, capable of carrying oxygen and life-saving targeted in the absence of blood. Firstly, superparamagnetic nanoparticles (MNPs) were synthesized, four different methods that are tartaricacid, glutaraldehyde, triethanolamine, 3-aminopropyltriethoxysilane were used for providing size stability of these materials. The surface of nanoparticles were gradually modified for connecting to organic molecules. Magnetic nanoparticles were characterized by FT-IR, elemental analysis, SEM, XRD, ICP, C-TEM. Pure hemoglobin molecules connected to surface of nanoparticles and has made repeated efforts characterization. Cyclic voltammetry and UV-Vis also used for invastigation of oxygen bolding-leaving ability of hemoglobine molecules. Hemoglobin bonded MNPs that mimic the natural erythrocyte were treated on the cell lines of endothelial cells culture to determine toxic effects of particles in vitro condition. 10 ml suspension of MNPs at definite concentration (1 mg/10 ml) treated on prepared cell lines at definite times (24 hours). Superoxide dismutase(SOD), catalase(CAT), glutathione peroxidase(GPx) activities and cell proliferation were investigated (n=8) for the determining the possible toxic effects of the cells. Currently, an artificial biomaterial alternative of blood was developed to use in preoperative and operative applications. It is thought that conducted this stage of study is a step for optimization/usability of synthesized hemoglobin-based solution and provide practice opportunities in different areas in future.
