Abstract (EN)

In this study, the biomolecular interaction occurred between nucleic acids and Capsaicin (CPS), the active compound in chilli peppers, which has been reported to have anti-carcinogenic properties, was investigated for the first time herein using disposable electrochemical biosensor. It is aimed to perform the interaction between CPS and double stranded DNA, cDNA from total RNA isolated from Huh7 human hepatocellular carcinoma cell line and PCR samples. The detection limit of DNA concentration range from 1 to 5 mg/mL was found to be 0.62 mg/mL. The effect of time-dependent manner from 1 min to 30 min on the interaction of CPS with nucleic acids was explored upon to the changes at guanine signal coming from double stranded DNA and cDNA as well as PCR samples. The surface confined interaction of CPS with different type of nucleic acids was investigated by disposable pencil graphite electrode (PGE) in combination with differential pulse voltammetry (DPV) for the first time in the literature. The interaction of CPS with double stranded DNA was also determined by agarose gel electrophoresis to confirm the interaction of CPS with PCR samples. In our study, electrochemical sensor technology has been developed for detection of CPS with DNA interaction that provides disposable low cost, reliable, and fast results. There has been much work on the development of personalized medical technology and the interaction of drugs, toxins, and chemical substances with nucleic acids. Electrochemical biosensors are the most common method used to investigate these interaction mechanisms. In this sense, electrochemical sensors can provide information to elicit the effects of DNA-targeted molecules on nucleic acids and offer new opportunities, particularly for the identification of effective treatments for cancer. Keywords: Capsaicin, double helix DNA, Huh7 human hepatocellular carcinoma cell line, disposable pencilpen graphite electrode, electrochemical biosensor, differential pulse voltammetry, agarose gel electrophoresis.
