Determination of AgNOR protein levels in the human prostat cancer cell line (PC3) applied with capsaicin

In this study; we planned to determine the effect of different doses of capsaicin, a component of red pepper, on AgNOR protein synthesis in the human prostate cancer cell line PC-3 (ATCC® CRL1435 ™). The PC-3 cell line, which is commercially available, is cultured in the Cell Culture Laboratory of Medical Faculty, Medical Genetics Department in Düzce University and the active cells in the logarithmic growth phase are divided into 5 groups. While no substance was applied to the first group of these cells, the other 4 groups including active cells were exposed with capsaicin at different doses (25uµ, 50uµ, 100uµ ve 200 uµ). Slides were prepared for each of these groups and Ag-NOR staining were performed. Then mean AgNOR number and total AgNOR area /nuclear area (TAA/NA) values were calculated in PC-3 cell line. As a result of our study, there was a statistically significant difference between the PC-3 cell line with no agent applied and the PC-3 cell line (25uµ, 50uµ, 100uµ ve 200 uµ) applied with capsaicin at different concentrations for both TAA/NA and mean AgNOR number (X2: 13,903 , p=0,009 and X2: 83,716 , p=0,000), respectively. When binary comparisons of PC-3 cell line groups that have been applied with capsaicin at different concentrations (25uµ, 50uµ, 100uµ ve 200 uµ) are performed, there was a statistically significant difference between the PCN-3 cell line applied with 25 uµ concentration and both of 50 uµ and 100 uµ concentration capsaicin for the mean AgNOR numbers but there was not a statistically significant difference among mean AgNOR numbers of the other treated groups. In addition, although there was a dose-related decrease among TAA/NA values of the administered groups, this increase was not statistically significant. This shows us that a certain amount of capsaicin has a protective effect against human prostate cancer and the dose concentration and degree of purity are important for this, also the AgNOR method can be used as an indirect marker to obtain knowledge about the treatment strategy and the prognosis of the disease.
