Abstract (EN)

Pseudomonas aeruginosa is a a human opportunistic pathogen, that colonizes biotic and abiotic surfaces. It's an important source of nasocomial infections in Turkey and the rest of the world.It has been shown that the interbacterial signaling system called "quorum sensing" is efficient in the regulation of virulence factor production.Bacteria use this system which works with exracellular signal molecules, to determine their local concentration.Pseudomonas aeruginosa is the most commonly used gram negative bacteria in the studies designed to explain quorum sensing systems.In pseudomonas aeruginosa, three quorum sensing systems known as "rhl", "las" and quinolon "PQS" have been identified.The determination of relationship between the host and Psedomonas aeruginosa exposed to subminimal inhibitor concentrations of antibiotics, has gained importance in planning the treatment of infections caused by this organism.In today's conditions, the determination of the genotype and phenotype of pseudomas species is as important as antibiotherapy in the treatment of infections caused by this type of bacteria.In this study the quorum sensing systems and biofilm production of las mutant Pseudomonas aeruginosa, rhl mutant Pseudomonas aeruginosa and standard Pseudomonas aeruginosa PAO1 strains were determined in vitro, in the presence of MIC and sub-MIC concentrations of imipenem, ertapenem, meropenem. Afterwards, HEp-2 cell cultures were infected with these strains in vitro, and nitrite and apoptosis-necrosis reactions were evaluated.As a result, we think that understanding the relationship between las and rhl mutant Pseudomonas aeruginosa strains and the host, the effect of quorum sensing systems on pathogenicity, and the role of quorum sensing in new/potential treatment methodologies will aid in the efficiency of treatment in Pseudomonas aeruginosa infections.
