Comparative pharmokinetics of carprofen oral, i̇ntravenous and subcutaneous administration in goats

Abstract (EN)

The aim of this study is to determine and compare pharmacokinetic profiles of oral, intravenous and subcutan administrations of carprofen, which is a nonsteroidal anti-inflammatory drug. Fifteen healthy Alphin goats, 12-14 months old and weighting 33-38 kg were used in this study. The animals were divided into three groups of 5 each such the equal mean weights. 1,4 mg/kgs carprofen were administrated orally to group I, intravenously to group II and subcutaneously to group III. Heparinized blood samples (5 ml) were collected from Vena vugularis of all animalsprior to drug administration and between 2nd minute and 120th hours after treatment. Concentrations of carprofen in plasma were measured by using high performance liquid chromatography. No adverse effect was observed during this study. After the administration of carprofen the drug concentration could be measured in plasma up to 120 th hours. The results indicated that intravenous administration produced a significantly higher maximum plasma concentration (11,92±0,76 µg/ml) as compared with subcutaneous (4,11±1,51 µg/ml) and oral administration (3,71±0,26 µg/ml). The time of maximum plasma concentration were shorter (5,00±2,45 hour vs. 7,00±1,22 hour) following subcutaneous administation compared with oral administration. The terminal half life was shorter in oral administration (27,43±1,78 hour) as compared with subcutaneous (33,93±7,48 hour) and intravenous administration (30,99±2,00 hour). Intravenous administration produced a significantly shorter mean residence time (34,54±1,35 hour) as compared with oral (37,09±3,85 hour) and subcutanousadministation (39,71±1.34 hour). In this study carprofen was administrated by different methods and the bioavailability of carprofen was determined greater in subcutanous administration as compared with other methods. If its aimed to obtain the maximum plasma concentration in minimum time intravenous, subcutanous and oral administrations will be effective respectively.
