Development of drug forms comprising antioxidant in the treatment of incision wounds

Abstract (EN)

The aim of this study was to formulate the mucoadhesive drug forms for intraoral application of glutathione with different excipients, characterization of them, conducting in vitro release study for the prepared formulations, studying the mucoadhesive and swelling characteristics of the formulation which was found to be suitable as a result of the release studies and evaluation of the healing effect of the selected formulation on the intraoral incision wounds of rabbits.The formulations comprising glutathione were prepared by the lyophilized or direct compression methods. The formulations to be prepared lyophilization were prepared by addition of sodium alginate and PEG 6000 or cyclodextrins of different types. Prepared gels were dried by lyophilization and the release specifications of the discs compressed from the obtained powders were examined. Various formulations were prepared by using two different alginates, two different chitosans, hydroxyprophylmethylcellulose, polycarbophil, two different poloxamers for the direct compression, and drug forms in the shape of discs were compressed from the powders.As a result of the in vitro release studies, the formulation comprising chitosan and glutathione was selected to use for in vivo experiments. Incision wounds were applied on gingival mucosa of the male New Zealand white rabbits, and the rabbits were separated into 4 groups. The rabbits of the first group had only incision wound (n=6). The rabbits of the second group were applied incision wound and then discs comprising GSH+chitosan were inserted therein. (n=6) The rabbits of the third group were applied incision wound and then discs comprising solely chitosan were inserted therein (n=5). The rabbits of the fourth group were applied incision wound and then discs comprising solely GSH were inserted therein (n=5). On the fifth day after the injury, oxidative parameters (MDA, GSH and NOx) in the scar tissues of rabbits were assayed. Tissue MDA levels in the group treated with the disc comprising GSH and chitosan were found to be lower compared to other groups. There was no statistically significant difference between the group treated with the disc comprising GSH-chitosan and the group having solely incision wound in terms of tissue GSH levels. Similarly, there was no statistically significant difference among the NOx levels.According to the hystological findings, wound healing in the group treated with the disc comprising solely chitosan was found better than the other groups.The results of the experiments showed that the application of prepared chitosan-GSH formulation and discs comprising solely chitosan on the intraoral incision wounds can be effective in wound healing.
