Abstract (EN)

Identification and detection of organic compounds at trace levels is an important analytical problem in biochemistry, medicine and environmental control. There is an increasing demand for simple, inexpensive and rapid analytical tests for the determination of trace concentrations of acetone in biological matrixes as a biomarker for diabetes mellitus. In this thesis, it was aimed to develop a chromatographic method for sensitive and selective acetone determination in gaseous and in synthetic urine for searching any potential to be applied to breath and real urine samples. Considering that acetone in biological matrixes is in trace level, an enrichment technique is necessary. In this study, solid phase micro extraction (SPME) fibers were used as the effective tools for sample pretreatment and enrichment. Gas chromatography systems equipped with a mass and flame ionization detectors were utilized for the quantification. Experimental parameters were optimized by either one-variable at time or factorial design. According the GC-FID results, adsorption time of 19.6 min, adsorption temperature of 49.8oC and pH of 2.14 was found as the optimal conditions. The LOD level calculated was found as 6.0 ng/mL and this method can be applied for a real sample.
