Abstract (EN)

As membres of a large superfamily of low-molecular-weight GTP-binding protein, ras proteins have played essential roles in controlling the activity of several crucial signaling pathways that regulate normal cellular proliferation. Activation mutations within the ras proteins frequently contribute to the development of human tumors.Collagens are the most abundant proteins in mammals. Collagen-rich extracellular matrices are not only critically impotant for the biomechanical properties of tissues, but are also intimately involved in cell adhesion and migration during growth, differentiation, morphogenesis and wound healing. The aim of this study was that determination of adhesion capacities of H-ras activite cells, 5RP7 to collogen type IV protein and also to find out how apoptotic process effected by adhering the cells to collagen type IV.The rate of 5RP7 cell growth was determined by trypan blue during the time points. The results showed that 12.5x104 cells/ml was optimum cell number for other experiments. The adhesive capacity of 5RP7 cells to various dilution of collagen type IV was investigated with either MTT assay or crystal violet method. The data obtaired from both methods showed that the cells had a higher adhesive capacity to even at 2.5 µg/ml collogen type IV compeared to control cells. Apoptotic DNA ladder was not appeared in 5RP7 cells adhered to collogen type IV by DNA fragmentation assay. The cells were bind to collogen type IV firmly.
