Abstract (EN)
SUMMARY Investigation of the Effects of Curcumin in Prevention of Colistin Nephropathy in Rats Objective- Hypothesis: Colistin is used to treat infections caused by multidrug-resistant gram-negative bacteria, which are becoming increasingly common. Antioxidant and anti-inflammatory agents have been tried to prevent effects such as nephrotoxicity and neurotoxicity, which limit the use of colistin. In this study, it was aimed to investigate the effects of curcumin in preventing colistin nephropathy. Method: Wistar albino rats weighing 300-500 g, without gender difference, were divided into 5 groups and included in the study. Group 1: (n = 8) control, group 2: (n = 6) colistin, group 3: (n = 7) curcumin + colistin, group 4: (n = 8) curcumin, group 5: (n = 8) olive oil were A total of 37 rats including examined. Groups were sacrificed after 7 days of treatment. Blood was collected from each rat into a biochemistry tube, and their 24-hour urine was collected in 50 ml falcon tubes. Kidneys were removed to study pathology and biochemistry. Research data were evaluated using SPSS 21.0 statistics program. p <0.05 was considered significant. Results: There was no statistically significant difference in serum cystatin-c, IL-6, levels between the curcumin + colistin group and the colistin group. The mean serum TNF-a was 178.5 ng / mL in the group receiving colistin and 120.6 ng / mL in the group receiving curcumin + colistin. When 24-hour urine volumes were examined, it was found that an average of 23.4 ml in the colistin group and 27.1 ml in the colistin + curcumin group. The creatinine clearance in 24-hour urine was 13.8 mL / min in the colistin group, while it was 16.1 mL / min in the colistin + curcumin group. When NGAL levels were evaluated in 24-hour urine, the mean value was found to be 13.8 ng / mL in only the colistin group and 12.5 ng / mL in the colistin + curcumin group. While there was no difference in TOS values of the two groups, when the TAOS values were examined, the mean value was 2.7 U / mL in the group receiving only colistin, and 1.7 U / mL in the group receiving colistin and curcumin. In pathological evaluation, there was no statistically significant difference in both groups. Conclusion: According to this study, some kidney damage occurred in the group receiving colistin, but it could not reach a statistically significant level. It was observed that oxidative stress, inflammation and kidney damage were less in the group with curcumin added. In the light of these findings, we think that curcumin is an important molecule that prevents colistin nephropathy. It is obvious that more studies are needed to reach the correct dose and duration.
