Analysis LNC-RNA crnde gene expression levels in colorectal cancer

Abstract (EN)
BACKGROUND: Long non-coding RNA (Lnc-RNA) has been found to be effective at various stages of cancer development with recent studies. Lnc-RNAs have been discovered to be important regulators in chromatin organization, transcriptional and post-transcriptional arrangements by interacting with DNA, RNA and proteins. The degraded gene expression of Lnc-RNAs is involved in cancer initiation, progression and metastasis. The aim of this study is to analyze the expression levels of the CRNDE gene, a Lnc-RNA in colorectal cancer, and to evaluate it clinically. MATERIALS AND METHODS: For this prospective study, approval was received from Gaziantep University Faculty of Medicine Local Ethics Committee with the decision of 2018/204 dated 26.09.2018. The study was conducted with the support of Gaziantep University Scientific Research Project (BAP) TF.UT.19.22. The study was carried out in 52 patients who underwent right hemicolectomy, left hemicolectomy, rectum anterior resection, low anterior resection, abdominoperineal resection in Gaziantep University General Surgery Department between 2014-2016. Lnc-RNA CRNDE gene expressions in each patient's tumor and adjacent non-tumor tissue were evaluated by Real Time PCR. Normalized ΔCt values were compared with Wilcoxon test. The coefficient difference between the gene expressions was compared with the 2-ΔΔCt method. RESULTS: The mean age of 52 patients operated on with colorectal cancer was 58, 1 ± 13, 1. On average, 80% of patients were stage 2-3. In real time PCR, CRNDE expression level ΔCt value in tumor tissue was found to be 7, 48 ± 3, 21. CRNDE ΔCt value in adjacent tumor-free tissue was 5, 57 ± 0, 95. It was determined that the expression levels of CRNDE in tumor tissue decreased approximately 5-fold compared to adjacent tumor-free tissue (by 2-ΔΔCt method). This expression difference increased as the stage of the disease increased. When only patients with rectal cancer were evaluated, it was seen that the level of gene expression in tumor tissue decreased by 8 times. DISCUSSION AND CONCLUSION: This study showed that CRNDE gene expression was significantly reduced in colorectal adenocarcinoma tissues. In contrast, many studies in the literature indicate that CRNDE expression is increased in colorectal cancer. Inconsistent results suggest that there may be other gene signal pathways that are not defined in CRNDE expression. In addition, the patient groups should be selected more homogeneously and their numbers should be increased sufficiently to be evaluated together with the relevant pathways and supported by cell cultures. We are of the opinion that it is not clinically sufficient alone in diagnosis and treatment. Key words: Colon, Rectum, Adenocarcinoma, Lnc-RNA, CRNDE, Gene expression
