ABSTRACT Investigation of Methylene Tethrahydrofolate Reductase Gene Polymorphism with Coronary Atherosclerosis Ethiyology at Coronary By-pass Patients Methylen tethrahydrofolate reductase (MTHFR) is an enzyme that catalyses the conversion of 5,10 methylen tethrahydrofolate to 5 methyl tethrahydofolate. MTHFR plays a role in the metabolism of folates which place in several biochemical pathways such as methylation of homocysteine and biosynthesis of nucleotides. MTHFR provides the balance between DNA synthesis and methylation reactions. Furthermore this enzyme also affects plasma homocysteine level which is a risk factor for cardiovascular diseases. MTHFR gene has been mapped to lp36.3. cDNA of human MTHFR gene isolated partially in 1994 and sequence of the cDNA was approximately 2,2 kb and consisted of 11 exon. The aim of this study was to investigate relationship between coronary atherosclerosis and MTHFR 677 C->T /1298 A-»C polymorphisms. For this purpose, areas where polymorphisms located in, was amplificated with Polymerase Chain Reaction (PCR) method. Polymorphisms were determined by using available restriction enzymes with Restriction Fragment Length Polymorphism (RFLP) method. Present study was progressed with control group. In this study, it was observed that there was no significant difference between MTHFR 677 C->T MTHFR, 1298 A->C and coranary atherosclerosis (for 677 C->T; p=0.240, for 1298 A->C; p=0.278), gender (for 677 C->T; p=0.961, for 1298 A->C için; p=0.980), the number of atherosclerotic arthers (for 677 C->T; p=0.181, for 1298 A- »C; p=0.747). It was found that there was no significant difference between cigarette/ alcohol intake and coronary atherosclerosis (for cigarette intake; p= 0.713, for alcohol intake; p=0.429).
