Abstract (EN)
In this study, all features of arginase in the sheep mammary tissue was been optimized in our laboratuar. It was determined that MnCk and preincubation were essential for the activation of the enzyme. It was found that preincubation temperature was 52°C at 12 minutes for arginase. The enzyme activity non-competitively was inhibited L- amino acids such as L- ornithine and L-lysine. EDTA, p- CMBA, NEM, methylene blue were significantly reduced arginase activity of the sheep mammary tissue which is in lactating periods. The enzyme activity was inhibited competitively by EDTA and non-competitively by the others. Arginase which has role in the destruction of arginin metabolism was localized in nucleus and has more activation at pH 9.5. It was found that methylene blue was effected on the arginase activation in different light. In addition, present study were demonstrated that, methylene blue, 150 watt light and MnCİ2 effects photoinactivation of arginase in the sheep mammary tissue. Km of arginase in the sheep mammary tissue were found 1.35mM. Because of arginase of the sheep mamary tissue has low Km such as ureothelic type.
