Özet (EN)

Ninety percent of Chronic myelocider leukemia cases have Philadelphia chromosome (Ph). Ph chromosome occurs with the translocation between 9th and 22 th chromosomes. BCR-ABL fusion gene occurs after this translocation (4). The aim of this study is to show bcr-abl hybrid gene existance with RT-PCR method in 25 CML cases and by estimating the Ph chromosome in cytogenetic way to determine the corelation betwen them. In our cases it's established to be found the ratio of bcr-abl gene positiveis 80% and the ratio of Ph chromosome positive is 80%. K562 cell line is used as positive control and pure water is used as negative controls of total RNA's.Total RNA's are isolated from blood, bone marrow and cell lineage; cDNA's synthesis from mRNA's so target DNA's are multiplied by PCR method. Oligonucleotide primers are applied to confrim of ordering bcr-abl in 25 CML cases. PCR product is performed in agorose gel. By dying the gel with ethidium bromide, screen is obtained under UV. light. In cytogenetic analysis lympocyte culture has been done from bone marrow material and by doing GTG dying microscope analysis has been done. It requires more further studies for understanding bcr-abl gene's intracellular functions of protein products and by using which mechanisms it transformes cells. As the results that we had, we understood that PCR method, which is more quick and more sensitive, could be used for diagnosing the malign illness and watching the response for the treatment.
