Synthesis, characterization, physical, chemical and biological properties of gold (AU) based hydroxyapatite biomaterials with quercetin and gallic acid additives

Abstract (EN)
In the presented master's thesis, the physical, chemical, biological, theoretical, and cytotoxic properties of Au-based HAp biomaterials containing gallic acid and quercetin produced using a wet chemical method were investigated. Characterization of the produced HAp biomaterials was carried out using X-ray diffraction (XRD), Fourier transform infrared (FT-IR), Raman spectroscopy, Scanning Electron Microscopy (SEM), Energy Dispersive X-ray (EDX), TGA/DTA, and DSC analyses. Furthermore, the theoretical properties of these biomaterials were determined using Hartree Fock (HF) and Density Functional Theory (DFT) methods. Finally, the in vitro cytotoxic properties of the produced HAp biomaterials were determined using the MTT assay against MG-63 human bone cancer cells, hFOB 1.19 human osteoblast cells, L929 mouse fibroblast cells, and Caco-2 human colon cancer cells. Analyses revealed that the Au, quercetin, and gallic acid-doped biomaterials exhibited the characteristic physical, chemical, biological, thermal, morphological, theoretical, and cytotoxic properties of pure HAp. All HAp samples were free of any chemical impurities and possessed the functional groups typical of HAp, with the Au element incorporated into the structure also being present in the chemical composition. Finally, the HAp biomaterials produced were found to have no cytotoxicity on the L929, MG-63, and hFOB 1.19 cell lines, and consequently, their biocompatibility was high.
