Abstract (EN)
In this study, the tannase enzyme from Lactobacillus plantarum and Lactobacillus brevis bacteria were produced and characterized. Tannase production of these bacteria were determined by production of clear zones around the colonies on the tannic acid containing medium, after 72 hours incubation at 35oC. Molecular weight of tannase from L.plantarum was estimated as 76.9 kDa on SDS-PAGE. The optimum activity of L.plantarum extracellular enzyme was emerged at 45oC, pH 6.0 respectively. Extracellular enzyme from L.plantarum was 78% stable for 5 minutes at 90oC. L.plantarum extracellular enzyme activity significantly stimulated with ZnCl2 (105%). Molecular weight of phytase from Lactobacillus brevis was detected as 120.9 kDa on SDS-PAGE. The optimum acitivity L.brevis extracellular enzyme was exhibited at 65oC, pH 8.0.Extracellular enzyme from L.brevis was 92% stable for 10 minutes at 90oC. L.brevis extracellular enzyme activity significantly stimulated with CaCl2 (%135) HgCl2 (%134) NiCl2 (%106). As a result, it is seen that due to the characteristic features of the L. plantarum and L.brevis bacteria extracellular tannase enzyme they can be proposed that they are convenient to be used in Industrial area.
