Acute toxicological effects of lavandula stoechas essansial oil and interaction of ephyleptic affect with antiephyleptic drugs

The first aim of this study was to determine the LD50 dose of Lavandula stoechas essential oil. The second aim of this study was to investigate ephileptojenic activities of the Lavandula stoechas essential oil.To find the LD50 dose, mice were divided into nine groups of six animals each. First group used as control group. And the other 8 groups were injected Lavandula stoechas essential oil i.p. respectively by the doses of 0.4ml/kg, 0.6ml/kg, 0.8ml/kg, 1.2ml/kg, 1.6ml/kg, 2.4ml/kg, 3.2ml/kg and 4.8ml/kg. After injections behavioral changes, convulsions, lethality and coma..etc. were observed. In the light of these findings LD50 dose determined as 1,88 mg/kg by probite analyse.To find the ephileptojenic activities of the essential oil we injected the oil and the antiephileptic drugs consicutively. We decided the Ls essential oil at the dose of 0.6 mg/kg which was caused convulsions but didn't kill the mice. At this part of the study, mice were divided into thirteen groups of ten animals each. First group used as control group. And the treated groups were given Lavandula stoechas essential oil i.p. 15 minutes before the administration of antiephileptic drugs. We used three diffrent doses of diazepam (1 mg/kg, 2 mg/kg, 4 mg/kg), fenitoin (10 mg/kg, 30 mg/kg, 50 mg/kg), fenobarbital (10 mg/kg, 20 mg/kg, 50 mg/kg) ve Na-valproat (100 mg/kg, 200 mg/kg, 400 mg/kg) as an antiepileptic drugs. After injections behavioral changes, convulsions, lethality and coma..etc. were observed. Starting time of changes which were observed before seizure, starting time of seizure, time of seizure and finishing time of seizure were determined. The statistical difference between the means of group and control was compared BY Mann-Whitney U test. Diazepam and fenobarbital have clearly prevented convulsions and lethality which were observed.As a result: LD50 dose of Ls determined as 1,88 mg/kg and proconvulsant effects were determined. Camphor, camphen and thujen which were exsisted in hydrodistillated oil could be responsible of these effects. Diazepam and fenobarbital could be used as antidote at toxication with Ls essential oil and also used for preventing toxication caused by Ls ess oil.
