Determinatin of the activity of leucojum aestivum extracts on antioxidant, antimikrobial and plasmid DNA

Abstract (EN)
The aim of this study was to evaluate the activity of Leucojum aestivum extracts antioxidant, antiMİKrobial, plasmid DNA and to determine the total phenolic compounds of Leucojum aestivum extracts by reverse phase high performance liquid chromatography. Leucojum aestivum is known as summer snowflake. Leucojum aestivum is categorized in the group of medicinal, aromatic and ornamental plants in Turkey. İn this study, extracts with etanol, metanol and hexane solvents were prepared to determine the potential of the plant to be separated by above ground and above ground parts of the plant. The antioxidant effects of the extracts were determined by 1,1-diphenyl-2-picryl-hydrazyl (DPPH), iron reduction, metal chelating, total flavonoid and total phenolic methods. Infrared spectroscopy of the extract was analyzed by FT-IR (Fourier Transform Infrared Spectroscopy). TLC (Thin Layer Chromatography) was used to determine functional groups. The results obtained were compared with the antioxidant effects of butylated hydroxyanisole (BHA), Gallik asit, kersetin, resorcinol and ascorbic acid. Total phenol content of the extracts were determined and peak areas were determined by HPLC analysis at different wavelengths. The amount of kateşin, kersetin flavonoids and Gallik asit phenolic component in extracts prepared by HPLC were determined. As a result, the etanol and metanol extracts of Leucojum aestivum show high antioxidant activity, including high phenolic and flavonoid. In addition, the etanol and metanol extracts of Leucojum aestivum showed antiMİKrobial activity against Gram-positive, Gram-negative bacteria and yeast and the results of antiMİKrobial activity were related to DNA interaction. According to the results of FT-IR analysis of the aboveground andsubsoil extracts, peaks were observed in aliphatic and aromatic regions. No NH group was found in subsoil extracts. According to the results of TLC, the components which have fluorescent properties were found in the aboveground extracts. In addition, the presence of flovonoid properties were found.
