Abstract (EN)

Lidocaine is the most commonly used local anesthetic containing the amide group. Lidocaine hydrochloride belongs to the group of narcotic drugs and can be used as a topical anesthetic that reduces the feeling of pain caused by the stability of the nerve membrane and the pain that occurs at the end of the nerve signals in the skin. Determination of lidocaine is very important because lidocaine overdose causes side effects such as drowsiness, visual disturbances, sweating, weakness, heavy breathing, muscle twitching, euphoria, agitation, attention deficit, speech difficulty, diplopia, hearing voices that do not exist, tremors, coma, chest pain, pale skin, ringing in the ears, difficulty swallowing, fast or slow heartbeat, loss of consciousness, hearing loss, seizure, unusual bleeding or bruising and vomiting. In this thesis, a PVC membrane lidocaine hydrochloride-selective potentiometric sensor (electrode) was developed using MIL-53 (Al) (Matériaux de l′Institut Lavoisier n o53), from metal organic frameworks as the ionophore. The optimum membrane composition was determined as 5% MIL-53 (Al), 62% DBF (dibutylphthalate), 32% PVC (polyvinyl chloride), 1% KTpClPB (potassium tetrakis(4-chlorophenyl)borate). The sensor prepared with optimum membrane composition exhibited a linear response in the concentration range of 1.0×10−7 molL-1-1.0×10−1 molL-1 with concentration change of 55.4 mV/decade and with a LOD (limit of detection) of 5.0×10−8 molL-1 for lidocaine hydrochloride prepared in deionized water. The pH working range of the sensor is determined as 2.37-7.45. The response time of the sensor was determined as 10 seconds. These performance characteristics indicated that the potentiometric lidocaine hydrochloride-selective potentiometric sensor had quite superior properties. The potentiometric application of the lidocaine hydrochloride-selective potentiometric sensor was successfully applied in pharmaceutical preparations.
