Abstract (EN)

Lipopolysaccharide (LPS) is an endotoxin found in the cell wall of Gram (-) bacteria, causing acute infection in living organisms. Although it threatens the vital functions of many organisms especially humans, LPS structure deactivates at very high temperatures. In this study, the damage caused by LPS in the tissues and the protective effect of vitamin E (VE) and sodium selenite (SS), which are known to be antioxidants, against these damages were examined on the lung tissues of rats. For this purpose, male rats were divided into 8 groups which are: the control group, SS exposed (0.35 mg/kg) group, vitamin E (VE) exposed group (200 mg/kg), VE+SS exposed group (200+0.35 mg/kg), LPS exposed group (10 mg/kg), LPS+SS (10+0.35 mg/kg) exposed group, LPS+VE exposed group (10+200 mg/kg), and LPS+SS+VE exposed group (10+0.35+200 mg/kg). At the end of the 6-hour period, antioxidant enzyme activities and damage in lung tissues were compared with the control group at Deoxyribo Nucleic Acid (DNA) level. As a result, the activity of antioxidant enzymes such as Superoxide dismutase (SOD), Glutathione peroxidase (GPx) and Catalase (CAT) in the LPS-exposed group decreased significantly compared to the LPS-exposed together with antioxidants groups. On the other hand, it was observed that the level of Malondialdehyde (MDA), known as an oxidative stress marker, decreased in antioxidant groups.. In addition, according to the results of the comet test, DNA damage caused by LPS was also observed. According to another result, DNA damage was found to be less in the group treated with antioxidants compared to the groups treated individually. As a result, diets in which different antioxidants are used together should be preferred to reduce LPS-induced tissue and DNA damage.
