The investigation of nesfatin-1, nitric oxide and ghrelin levels in major depressive patients before and after treatment

Abstract (EN)

In this study, some biochemical parameters such as cholesterol, LDL, HDL, tryglyseride, glucose; serum concentrations of acyle and desacyle ghrelin, nesfatin-1 and nitrik oxide have been measured in major depressive patients, taking Efexor, before and after treatment and with control groups, and its relation with ghrelin and nesfatin-1?s correlation of weight gain and relationship of nitric oxide are determined.Hunger blood samples were taken from twenty major depressive patients before and after their treatments that lasted two months and also blood samples were taken from 40 healthy subjects whose BMI were matched with these patients?. 225 mg Efexor were given to major depressive patients once a day for two months.Blood ghrelin, acyle and desacyle, and nesfatin-1 concentration were measured using ELISA method. Nitric oxide levels were determined spectrophotometrecally using by the Griss reaction. There was a low but significant change in acyle ghrelin and desacyle ghrelin levels; before and after treatment group compared to the control group. However, there was a low but a non- significant change in the group after treatment compared to before treatment.Nesfatin-1 was measured for the first time in patients with major depressive. Despite nesfatin-1 level was determined to be highest before treatment, but it declined with treatment and it was still remarkably higher than the healthy subjects. There was high and significant change in nesfatin-1 level, before and after treatment group, compared with control group; and low but significant change found in after treatment patient group compared before treatment.Nitric oxide level was found to be lowest before treatment, and despite increase after treatment it was found significantly lower than the healthy subjects. While there was significantly low value in before and after treatment patient group compared to the to control, it was found high but not significantly change in after treatment patient group compared before treatment patient group.Cholesterol, LDL, tryglyseride and glucoz levels were found to be highest in after treatment patient group and were found lowest in control group. The highest HDL level was found in control group, lowest in before treatment patient group.These data shows that decreased ghrelin and increased nesfatin-1 level can be related to gaining weight in major depressive patients and can be used as two efficient parameters in the diagnosis of this illness.As a result, when chosing an appropriate drug therapy for these patient groups, it is extremely important to lead a decrease in ghrelin and nesfatin-1 and increase in nitric oxide levels, which can cause a markedly high level of recovery.Key words: Depression, Nesfatin-1, Nitric Oxide, Ghrelin, Efexor, Venlafaksin.
