Abstract (EN)
Introduction; Mantle cell lymphoma (MCL) is aggressive sub-type of Non- Hodkgin Lymphoma which is characterized by expression of cyclin D1. It is not a curable disease with available chemotherapeutic regimens in nowadays. Ideal therapeutic investigations are performed for this reason. These investigations were shown that cell cycle is deregulated in the cells and targeted therapies directed to phase of cell cycle may cause important development of therapies in MCL treatment. The cell cycle regulators p53, p63, Rho kinases play an important role in the pathogenesis of cancer has been reported to be. Again studies Ki-67 proliferation index was shown to be an important prognostic factor in cancer patients . Aim; in this study, we thought that cyclin D1 and p53, p63 and Rho-kinase (ROCK1 and ROCK2) enzymes may be an association of, MCL. We planned to investigate and compare the protein expression profile cyclin D1, p53, p63, and with the ROCK1, ROCK2, and also ROCK2 gene ROCK1 polymorphisms and Ki-67 expression in patients with MCL. We also aimed to investigate response to treatment, the relationship between recurrence and prognosis. Material and Method; This study was performed retrospectively and was included 60 MCL and 60 control group of patients who applied to hematology department of Gaziantep University of Medical School between 2006-2012 years. Hematoxylin and eosin stained slides in the entire archive was reevaluated and selected for immunohistochemistry and PCR again. p53, p63, ROCK1 and ROCK2 were scored to between 0-3 rates according to staining ratio in tumor cells. Ki-67 was expressed as % according to staining of tumor cells Result; There was significant increase between ROCK1 (P=0.0009) and ROCK2 expression in MCL patients compared with control group (P<0.0001). ROCK2 expression was higher than ROCK1 in MCL patients (P<0.0001). There were significant positive correlations between age and ROCK1 p (p=0,044). Expression of p53 and Ki-67 were stronger in patient group than control group. There was no polymorphism detection in the analysis of ROCK1 gene polymorphism in patients and control groups. There was no significant relation in ROCK2 polymorphism in gene polymorphisms analysis between in MCL patients and control group. Key words: Mantle cell lymphoma, ROCK1, ROCK2
