Abstract (EN)

Breast cancer, which is the most common cancer type in the world, takes the second place after lung cancer in our country. A good understanding of the features of this pathogenesis plays an important role in the development of new treatment methods in breast cancer. In recent years, "Human Epidermal Growth Factor Receptor-2" (HER2) status, estrogen receptor (ER), progesterone receptor (PR) levels have been important prognostic factors. Combination therapy is the way in which two or more drugs or methods are used that has gained momentum in recent years. One of the most important aspects of this treatment is that it reduces the development of drug resistance, as the tumor or a pathogen will be less likely to resist more than one drug at the same time. Tamoxifen, an estrogen blocking and non-steroid compound, has been used to treat patients with hormone receptor positive breast cancer for nearly 20 years. Deinoxanthin is a xanthine derivative isolated from the S-layer in the cell wall of the radiation-resistant bacteria Deinococcus radiodurans. Studies have shown that this xanthine derivative shows pro-apoptotic and anti-proliferative effects on some types of cancer. Within the scope of the thesis, the anti-cancer and anti-proliferative synergistic effects of Tamoxifen and Deinoxantin on the MDA-MB-453 breast cancer cell line were investigated. In this context, as a result of applying single and combinations of substances to cell culture; Investigation of expression levels and protein levels of Bax, Bcl2, Caspase 3 and HER2 genes were carried out by RT-qPCR and ELISA methods.
