Abstract (EN)
In this thesis, microbial production of lycopene in Escherichia coli bacteria by metabolic engineering approach was aimed. For the microbial production of lycopene in E. coli, the plasmid (pAC-EHER-EBIidi) containing the expression of the enzymes that carry out the synthesis of lycopene using the Gibson assembly method (crtEBI) was carried out. The plasmid that was constructed had different E. coli strains (E. coli MG1655, E. coli NEB 10-beta, E. coli DH5α, E. coli DH5α-Pro, E. coli NEB-Turbo, E. coli XL1-Blue, E. coli TOP10, E. coli EPI300, E. coli DE3) was transferred and the liquid cultures of these strains transferred to the plasmid were prepared. The lycopene production from the prepared liquid cultures was carried out by means of acetople extraction with acetone and analysis of the extraction products by High Pressure Liquid Chromatography (HPLC)
