Investigation of serum, liver and adipose tissues desnutrin with chemerin expressions in rats with ınduced metabolic syndrome

Abstract (EN)

The metabolic syndrome (MetS) with an ever increasing prevalence around the world is a public health problem directly related to diabetes and cardiovascular system diseases. Although it is asserted that there are various factors such as inflammation, insulin resistance, dyslipidaemia, and peptide hormone in MetS etiopathogenesis, the underlying mechanisms have not been fully clarified yet. Therefore, in this study we aimed to investigate how the expressions of adipose triglyceride lipase (Desnutrin/ATGL) and chemerin hormones change and how the reflections of this change in the systemic circulation are in liver and adipose tissues of rats with MetS. In this study 14 eight weeks of age Sprague Dawley rats with a weight of about 260-290 gr were used. Whilst rats were being fed ad libitum with standard rodent chow and 10% fructose added to the potable water during the experiment, the rats in control group were given only rodent chow. Routine biochemical parameters were measured by using autoanalyzer. Levels of chemerin, Desnutrin/ATGL and insulin of tissues and serums were measured by using enzyme-linked immunosorbant assay (ELISA) technique. HOMA-IR levels were calculated benefiting from formula. Whilst levels of serum glucose, ALT, AST, triglyceride, LDL-K, total cholesterol, insulin, HOMA-IR and chemerin are high (P <0.05) in a statistically significant way, levels of HDL-K and Desnutrin/ATGL were determined low (P <0.05) in a significant way compared to the control group. Furthermore, it was determined that the expressions of Destunrin/ATGL in liver and adipose tissues of rats in MetS group decreased in a statistically significant way compared to the control group (P <0.05), the chemerin levels of liver and adipose tissue increased in a statistically significant way (P <0.05). Consequently, MetS causes chemerin amounts to increase, Desnutrin/ATGL amounts to decrease. Therefore, we consider that these two hormones take part in pathogenesis of the MetS directly and these findings can help to clarify the mechanisms underlying the MetS.
