Microbial hyaluronic acid production and investigation of the evaluability of agricultural waste in hyaluronic acid production

Abstract (EN)

Hyaluronic acid (HA) is a commercially valuable polymer bonded by β-1,3 and β- 1,4 glycosidic bonds and performs some vital activities in the living body. In this study, it is aimed to investigate different microorganisms that can be used in HA production, to evaluate the usability of agricultural wastes in fermentation environment and to optimize hyaluronic acid production conditions. S.thermophilus, S. epidermidis, E. durans, E. gallinarum, S. mutans and S. warnei were examined in terms of HA production. Optimization studies continued with the bacteria that were determined to produce HA through screening experiments. After determination to the factors affecting production with the Plackett Burman experimental design, the optimum production conditions were determined by the response surface method. The most important factors affecting the production were found to be temperature, pH, agitation speed and trace minerals The highest production amounts were found as S. thermophilus P4 88,0 mg/L and S. mutans 1a21 85,0 mg/L. After determination to the optimum production conditions, the usability of whey, molasses, chill and olive pulp in production was investigated. The hyaluronic acid produced has been determined by hyaluronidase enzyme and FTIR. It has been determined that the bacteria taken in the experiments can be used in the production of HA, and the evaluated agricultural wastes can be used as a carbon and nitrogen source in the HA medium.
