Abstract (EN)

MicroRNAs have got 20-30 nücleotid and they are RNA molecules that make up the class of non-protein-coding endogenous small RNAs. They have been proven by a lot of research they took charge as an oncogenic or tumor suppressor for a lot of cancer. These miRNAs also contribute to many biological processes such as cell regulation, development, apoptosis, and metastasis. Many such properties make the study on microRNAs interesting. Many miRNAs have been researched before, but there is not much research on miR-105-5p, which is a member of the miR-105 family. The purpose of this research; To examine the effect of miR-105-5p on lung (A549), breast (MCF7), and cervical (HeLa) cancers. For this purpose, the knockdown and over-expression conditions of miR-105-5p were provided by using the transfection method. Cells under these conditions; The growth rates were observed by establishing the proliferation experiment and the colony-formation characteristics were observed with the colony-formation experiment. In addition, the effects of miR-105-5p on apoptosis and stem cell-related genes in these cancers were also examined by RT-PCR and it was observed that these genes were affected as an oncogene or tumor suppressors. In addition, the increased expression of miR-105-5p suppressed the growth rate and colony formation properties of cells in all three cell lines. Based on these results, it is thought that miR-105-5p can be used as a targeted therapy.
