Investigation of the effect of mirabegron on pulmonary hypertension

In this study, we investigated whether Mirabegron, a selective beta-3 adrenergic receptor agonist currently used in overactive bladder, changes the course of the disease in experimental pulmonary arterial hypertension (PAH) rat model. In the study, it was aimed to show the effect of Mirabegron in reducing right ventricular enlargement, pulmonary artery thickening and pressure in subjects with pumonary arterial hypertension. Sprague Dawley rats between 230-470 g were used. Study groups Group1: control n=7; Group 2: Monocrotalin (MCT) 14 days n=10; Group 3: Monocrotalin (MCT) 28 days n = 10; Group 4: MCT 14 + Miragebron 14 days n=10; Group 5 MCT 28 + Mirabegron 14 days n=10. The control group did not receive any therapy, and the right ventricle pressure, carotid artery pressure were measured, and then the heart and lung were removed and the right ventricle / (left ventricle + septum) weights (Fulton Index) of the heart were measured. Pulmonary artery thickening and occlusion percentage were measured in samples taken from the left lobe of the lung. The parameters to be examined were examined after 14 days with a single dose of Monocrotalin in Group 2 and 28 days after Monocrotalin was administered in Group 3. Group 4 was treated with Mirabegron for 14 days after 14 days of Monocrotalin administration, and group 5 was treated with Mirabegron for 14 days after Monocrotalin administration, and parameters to be examined were examined. The right ventricular pressures of the groups were 6.67 ± 2.12, 6.13 ± 0.83, 14.95 ± 5.23, 6.23 ± 1.86 and 12.66 ± 4.88, respectively, and with p <0.05, group 3 was significantly higher than group 1, and there was no significant difference in the others. Pulmonary artery vascular thickening of the groups respectively; It was 4.22 ± 0.82, 9.85 ± 0.90, 14.59 ± 0.56, 11.81 ± 0.56 and 11.49 ± 0.73, and significantly (p <0.05) less vascular thickening was found in group 5 compared to group 4. It can be concluded that Mirabegron has a positive effect on improving PAH symptoms, but it appears that both animal and human studies are needed to examine broader parameters.
