In-vitro investigation of the effect of morusin on matrix metalloproteinase-1 in human fibroblast cell line

Abstract (EN)

Objective: To investigate the effect of morusin on MMP-1 in human fibroblast Hs68 cell line in-vitro. Method: Hs68 cells were multiplied and lifted with Trypsin-EDTA and divided into 96-well plates. At the end of the 24-hour incubation period, morusin was applied to the cells at a concentration range of 32-0.015625 μM/mL. Its effects on cell proliferation at the end of 24, 48 and 72 hours incubation was studied with the WST-1 kit. Proliferated cells were divided into 6-well plates this time. At the end of the 24 hours incubation period, morusin was applied to the cells at doses in the concentration range of 4-0.5 μM/mL. The effect on the MMP-1 release at the end of the 24, 48, and 72 hours incubation periods was studied with the MMP-1 kit and the absorbance values were taken with the ELISA Reader. Results: It was observed that Morusin killed almost all of them on the Hs68 cell line applied at concentrations of 32, 16 and 8 μM/mL. It was determined that it had a significant cytotoxic effect at 4 and 2 μM/mL concentrations, and less cytotoxic effect at 1 and 0.5 μM/mL concentrations. The amount of MMP-1 was increased 1.64% at 4 μM/mL and 1.72% at 0.5 μM/mL depending on morusin concentration in as the 24 hours incubation period, it decreased 2.80% at 2 μM/mL and 8.70% at 1 μM/mL. The amount of MMP-1 was decreased 1.90% at 4 μM/mL, 11.90% at 2 μM/mL and 4.12%, at 0.5 μM/mL in as the 48 hours incubation period, it was increased 1.67% at 1 μM/mL. In the 72-hour, the amount of MMP-1 was increased 10.64% at 4 μM/mL, 8.26% at 2 μM/mL; 21.78% at 1 μM/mL and 12.80% at 0.5 μM/ mL. Conclusion: The applied concentrations of morusin showed cytotoxic or cell proliferation reducing properties for Hs68 cells. It was observed that the morusin used decreased the amount of MMP-1 in only 2 concentrations depending on time, while it showed an antiaging effect for the cells, while other concentrations showed an accelerated aging effect.
