Determination of sirtuin 7, SEMA 3A, SEMA 3F gene expressions in patients with multiple sclerosis

Abstract (EN)

In this study, we aimed to contribute to the pathogenesis of the disease by studying SIRT7, SEMA3A and SEMA3F gene expression levels in the cerebrospinal fluid (CSF) and blood samples of multiple sclerozis (MS) patients. SIRTUIN7 (SIRT7) can help weaken DNA damage and therefore increase cellular survival under genetic stress conditions. In some cancer patients, SIRT7 is highly expressed, suggesting it may be an oncogene. Semaphorins are a family of intercellular signal proteins, most of which act as axonal guide molecules. Semaphorins are the most important oligodendrocyte progenitor cell (OPC) guide molecules. Patients diagnosed with MS as the disease group and control group without MS disease were selected and ACTB, SEMA3A, SEMA3F and SIRT7 gene expression levels were studied by performing total RNA isolation from CSF and blood samples. The study included 59 volunteers as 31 patients and 28 control groups. Gene expression levels were obtained by RT-PCR method using SIRT7, SEMA3A and SEMA3F genes from RNA samples obtained from Cerebrospinal Fluid and blood samples, and ACTB gene for endogenous control. When the SIRT7 gene expression levels in the blood samples taken from the patient and control groups were compared, 1.86-fold increase was observed in the patient group (up-regulated). When CSF samples were compared, it was found that there was a 5-fold increase (up-regulated) in SIRT7 gene expression in the patient group, a 36-fold increase in SEMA3A and a 17-fold increase in SEMA3F. Although gene expression levels in the blood samples of the patient group, 161 times increase in SEMA3A (up regulated) and 69 times increase in SEMA3F, it was found down-regulated compared to the control group. In this study, a significant increase was observed in the gene expression levels of SIRT7, SEMA3A and SEMA3F in MS patients, with a more pronounced blood sample. As a result, the expression levels of these genes can be monitored in samples to be taken from the blood sample without the need for a CSF sample, and the progression of the disease can be followed. However, larger studies are needed to reveal the relationship of SIRT7, SEMA3A and SEMA3F gene functions with MS. Keywords: Multipl Sclerosis, Gene Expression, Semaphorin, Sirtuin
