Effects on somatic embryogenesis of different growth regulators and different explant resources in pomegranate (Punica granatum)

Abstract (EN)

In this thesis, the effect of pomegranate varieties (Hicaz and Silifke Aşısı), different explant types (leave, hypocotyl, cotyledon, root explant) and different growth regulator combinations 2,4-dichloro phenoxy acetic acid (2,4-D) and benzyl adenine (BA) on somatic embryogenesis were investigated. In addition to plant growth regulators, the effect of poliamine derivative, spermine on explant regeneration were determined. The results have indicated that white callus formation in Silifke Aşısı (36.60 %) gave beter results than that of Hicaz variety (32.58 %). No statistical differences were obtained between two varieties on brown-black callus and embryogenic callus formation.Putative embryogenic calluses were obtained from the explants; cotyledone (10.1 %), hypocotyl (9.78 %), root (8.53 %) and leave (7.53 %) respectively.
